Título: Bioactive Scaffolds for mesenchymal stem cell therapy for bone regeneration

Resumo: Bone is a specialized connective tissue susceptible to pathological processes that can result in extensive bone defects, challenging to regenerate. Bone regeneration based on stem cell therapies has been investigated. This study aimed to develop scaffolds, with poly-L-lactide (PLLA) functionalized with hydroxyapatite (HA) and/or dicalcium phosphate dihydrate (DCPD) or by magnesium (Mg) and Strontium (Sr). Periodontal ligament stem cells (hPDLSC) were isolated and characterized to evaluate the osteoinductive and osteoconductive potential of scaffolds. Cell adhesion (SEM), proliferation (CCK-8 assay) at 3 and 7 days, and differentiation (alizarin) were evaluated. Data were submitted to ANOVA and Tukey. HA and DCPD were synthesized with ionic substitution with Mg+2, Sr+2, X-ray diffraction analysis confirmed calcium phosphate synthesis. Scanning electron microscopy (SEM) showed HA crystals with nanometric size and DCPD with plate-shaped microparticles. At 7 days, PLLA_HA_Mg_Sr demonstrated covered surface, while PLLA_DCPD_Mg_Sr showed spaces between the cells. The addition of HA increased cell viability and proliferation compared to PLLA. HA-based scaffolds demonstrated greater osteoinductive properties. Osteoinduction in PLLA_HA_Mg_Sr and PLLA_DCPD_Mg was greater, increasing the mineralization of the extracellular matrix in the osteogenic environment. The incorporation of magnesium and strontium with hydroxyapatite allowed cell adhesion, proliferation, and differentiation and could promote bone regeneration.
