Resumo: Objective: This review aimed to highlight the implications of DNA ploidy in head and neck squamous cell carcinoma (HNSCC), particularly its predictive value, role in patient stratification, and potential as a therapeutic target for this malignancy. Study design: A narrative review of the literature was carried out by searching electronic databases. Results: In conventional HNSCC, aneuploid tumors correlate with increased lymph node metastasis, locoregional recurrences, poor response to radiotherapy and chemotherapy, and worse prognosis. Few studies address the role of DNA ploidy in young patients with HNSCC. The results showed that these patients have high DNA ploidy abnormalities, indicating significant genomic instability and probably genetic differences compared to older patients. Regarding the association between HPV and DNA ploidy status, there was no difference between the DNA ploidy status of HPV-associated and HPV-independent tumors. About the role of DNA ploidy in histological subtypes, surgical margins, and target therapy, more research is needed on these topics. Conclusion: Here, we review the DNA ploidy findings in HNSCC. DNA ploidy seems to be useful in stratifying patients and predicting the outcome of HNSCC. Further research into new patient profiles, histopathological subtypes, surgical margins, and targeted therapies based on chromosomal instability is warranted. Support: CAPES
