Título: DIAGNOSTIC ACCURACY OF PLASMA-BASED LIQUID BIOPSY FOR DETECTING THE BRAF V600E MUTATION IN PATIENTS WITH AMELOBLASTOMA

Resumo: Objective: This study aimed to assess the diagnostic accuracy of plasma-based liquid biopsy for detecting the BRAF V600E mutation in circulating cell-free DNA (cfDNA) from patients with ameloblastoma. Study Design: Following STARD guidelines, we conducted a diagnostic accuracy study. The index test was plasma-based liquid biopsy, with conventional tissue biopsy serving as the reference standard, while the target condition was the presence of the BRAF V600E mutation. Individuals diagnosed with mandibular ameloblastoma were enrolled. Tumor tissue DNA and plasma cfDNA were isolated and subjected to quantitative real-time Polymerase Chain Reaction (qPCR) mutation detection assays utilizing allele-specific TaqMan probes. Sensitivity and specificity were calculated. Results: Twelve individuals with conventional ameloblastoma and three with confirmed wild-type BRAF lesions other than ameloblastoma (negative controls) were included. While the mutation was detected in 11 (91.66%) ameloblastoma samples, it was not detectable in any plasma cfDNA samples, both in ameloblastomas and the negative control group. Sensitivity and specificity measures for plasma-based liquid biopsy in detecting the BRAF V600E mutation were 0.0 and 1.0, respectively. Conclusion: Plasma-based liquid biopsy appears to lack accuracy in detecting the BRAF V600E mutation in circulating cfDNA from ameloblastoma patients, irrespective of clinicopathological features of the tumor.
