Título: ADENOID CYSTIC CARCINOMA INTERACTOME: EXPLORING MYB AND MYB-NFIB PROTEIN INTERACTIONS WITH TURBOID

Resumo: Objective: The aim of this study was to elucidate the protein-protein interactions associated with MYB and MYB-NFIB, to better understand the biological processes involved in adenoid cystic carcinoma. Methods: Human embryonic kidney cells (HEK293) were transfected with three distinct constructs: TurboID empty vector (EV), TurboID MYB, and TurboID MYB-NFIB. Untransfected cells were used as experimental control. Twenty-four hours after transfection cells were treated with 0.5mM Biotin for 20 minutes. MYB production and protein biotinylation were confirmed through western blot and immunofluorescence. Protein lysates were purified using streptavidin magnetic beads. Mass spectrometry analysis was performed on four independent biological replicates. Results: Mass spectrometry revealed distinct protein interaction profiles for both MYB and MYB-NFIB compared to the EV. In the MYB versus EV comparison, 16 proteins were up-regulated, while 550 were down-regulated. Similarly, the MYB-NFIB versus EV analysis showed 10 up-regulated and 336 down-regulated proteins. Despite variances from the empty vector, pairwise comparison between MYB and MYB-NFIB failed to disclose any statistically significant differences. Conclusion: This comprehensive exploration sheds light on the network of protein-protein interactions orchestrated by MYB and MYB-NFIB, providing valuable insights into the underlying biology of adenoid cystic carcinomas driven by the fusion protein.
