Título: CINNAMALDEHYDE AND α-TOCOPHEROL- LOADED NANOCAPSULES WITH CHITOSAN FOCUSING ORAL MUCOSITIS

Resumo: Objectives: In this work, we aimed to develop a therapeutic product containing cinnamaldehyde (CNM) and α-tocopherol (α-toc)-loaded nanocapsules cross-linked with chitosan to antimicrobial activity for oral mucositis application. Methodology: The active compounds were characterized by gas chromatography coupled to mass spectrometry and in silico through molecular docking. CNM/α-toc-loaded nanocapsules cross-linked with chitosan were produced by alternating cycles of shaking and cooling, and characterized by particle size, polydispersity index, zeta potential (ZP), scanning electron microscopy, FTIR, sol-gel phase studies, stability, rheology, antimicrobial activity, cell viability and antitumor activity in vitro. Results: Molecular docking study of α-toc and CNM for the inflammatory pathways indicated affinity for the TLR4 receptor involved in the regulation of cyclooxygenase-2 (COX-2) and nuclear factor-κB (NF-κB). They were stable, showing neutral ZP, low porosity, pseudoplastic behavior and sol-gel transition at body temperature. Antimicrobial activity against strains of Escherichia coli, Staphylococcus aureus and Candida albicans (MIC starting at 3.12 μg/mL) was enhanced when compared to non-encapsulated CNM. There was a trend towards selective toxicity, with a reduction in cytotoxicity and a gain in the antitumor activity of the encapsulated ingredients. Conclusion: CNM/α-toc-loaded nanocapsules cross-linked with chitosan showed good antimicrobial characteristics representing a potential formulation to treat oral OM.
