Resumo: Objectives: The aim of this study was to evaluate the early and late, clinical and histological, changes associated with the polycaprolactone (PCL) dermal filler in the orofacial region of rats. Study Design: Forty-eight female Wistar rats were divided into PCL and control groups. The material was injected into the ventral tongue and submandibular region. The animals were euthanized after 24 hours, 30 and 90 days. Results: The PCL group presented nodules on the tongue at all experimental times. Histological analysis, within 24 hours, showed the presence of the material and an intense inflammatory process. At 30 and 90 days, there was a predominance of macrophages and multinucleated giant cells around the PCL spheres. Dermal collagen density was higher in the PCL group when compared to the control at 30 and 90 days. In the submandibular glands, we observed an inflammatory process similar to that in other sites, without acinar or ductal morphological changes. Conclusion: The present study demonstrated the significant volumizing capacity of the material, initially maintained by the carboxymethylcellulose gel and then by the foreign body reaction and collagen neoformation. The formation of nodular lesions on the tongue warns of the risk of this complication in mobile anatomical structures.
