Título: ANALYSIS OF BRAF p.V600E MUTATION BY ddPCR IN PLASMA OF PATIENTS WITH AMELOBLASTOMA

Resumo: Objective: To investigate whether tumor DNA could reach blood circulation and, consequently, if cell-free DNA (cfDNA) could be detected by liquid biopsy in the plasma of patients with ameloblastoma. Study design: Peripheral blood samples and tumor tissue were collected from five patients. Mutational status analysis for BRAF p.V600E mutation was performed on gDNA (tumor tissue) and cfDNA (plasma) using allele-specific real-time qPCR and droplet-digital PCR (ddPCR), respectively, in the selected cases. Results: BRAF p.V600E was detected in four (4/5) ameloblastoma tissue samples, while all tested ameloblastoma plasmas were negative for the detection of BRAF p.V600E mutation in cfDNA. Conclusion: These preliminary findings suggest that with the current technology, BRAF p. V6000E mutation cannot be detected in ameloblastoma patients' plasma.
