Título: TOXICITY OF PHOTODYNAMIC AND SONODYNAMIC THERAPIES MEDIATED BY 5-ALA ON DYSPLASTIC ORAL KERATINOCYTES: AN IN VITRO STUDY

Resumo: Objective: To demonstrate the toxicity and morphological aspects of cell death induced by photodynamic therapy (PDT), sonodynamic therapy (SDT) and sono-photodynamic therapy (SPDT) mediated by 5-aminolevulinic acid (5-ALA) on dysplastic oral keratinocytes. Study design: Dysplastic oral keratinocytes (DOK) cell line was seeded into 35mm Petri dishes. 24h after treatment with PDT, SDT or SPDT mediated by 5-ALA, cell viability was measured by the tetrazolium salt (MTS) colorimetric method, and cell morphology was analyzed using an inverted phase contrast microscope. Results: The viability of DOK cells treated with 5-ALA, LED, or US alone was not significantly reduced in relation to the control group. Treatment with PDT, SDT, and SPDT reduced the cell viability significantly in relation to the control group. Morphological alterations after treatments were characterized by karyolysis, karyorrhexis, vesicles formation inside the nuclei, and formation of cytoplasmatic projections suggestive of pore formation in the cytoplasmic membrane. Conclusion: DOK cells are sensitive to PDT, SDT, and SPDT mediated by 5-ALA through different cell death mechanisms, suggesting that these methods represent potential therapeutic approaches for oral epithelial dysplasia.
