Resumo: Aim: to assess the gene expression of SARS-CoV-2 cellular entry factors ACE2, TMPRSS2 and FURIN in pleomorphic adenoma in comparison to normal salivary glands. Methods: Twenty-five formalin-fixed paraffin embedded samples were obtained, consisting of 15 pleomorphic adenoma, and 10 normal salivary glands. RNA extraction and RT-qPCR were performed to assess mRNA expression of ACE2, TMPRSS2 and FURIN. The results of target transcripts were normalized to endogenous levels. Results: mRNA expressions of ACE2, TMPRSS2 and FURIN were higher in normal salivary glands compared to the levels in pleomorphic adenoma (p<0.05). A positive correlation was observed between ACE2, TMPRSS2, and FURIN in pleomorphic adenoma (p<0.05). Conclusion: SARS-CoV-2 cellular entry factors are expressed in salivary glands. Our analysis revealed higher levels in normal glands when compared to the expression in salivary glands affected by a salivary gland tumor. Pleomorphic adenoma may downregulate the expression SARS-CoV-2 entry factors in the salivary glands analyzed. SUPPORT: FAPEMIG, CAPES
