Resumo: AIM: Evaluation of extracellular matrix (ECM) role in the tumor microenvironment (TME) in order to search for new strategies for target-cancer treatments. STUDY DESIGN: Literature searching guided by the keywords "tumor microenvironment", "cancer treatment", "extracellular matrix", and "target cell treatment" was made in the PubMed database, without publish date limitation. RESULTS: TME is a complex system, composed by ECM, vascular microenvironment, immune microenvironment and other cells. Extracellular matrix is responsible for coordinate cell behavior, and its dysregulation can improve cancer. ECM is composed of proteins, such as cancer-associated fibroblasts, elastin, fibronectin and collagen, growth factors and cytokines, which can regulate immune, stromal and cancer cells behavior. For example, collagen is a key factor in ECM, which undergoes most changes in tumorigenesis. Furthermore, ECM deposition leads to tissue stiffness, a main point in the tumorigenesis process, as it can lead to tumor advance. Modifications in ECM structure also assist in cell proliferation and metastasis. CONCLUSION: Considering that ECM cells and functions are essencial for cancer development and progression, knowledge of them in the TME are crutial for searching new strategies to promote target-cancer treatments without damaging normal cells.
