Título: L-GLUTAMINE IMPROVES TISSUE REPAIR OF ORAL MUCOSITIS IN ANIMALS TREATED WITH 5-FLUOURACIL

Resumo: Objective: To investigate the effect of application of L-Glutamine on tissue repair in oral mucositis in animals treated with 5-Fluouracil (5-FU).
Study design: C57BL/6 mice were used, divided into 2 groups (n=15/group), which received intraperitoneal injections of 5-FU for 3 non-subsequent days, at concentrations of 60mg/kg, 40mg/kg and 40mg/kg. Then, oral mucositis was induced with 5µL of 5% acetic acid on the belly tongue. One group was treated with 50 µL of L-Glutamine, twice a day at a concentration of 0.5g/kg, and the control received the same volume of filtered water, both for 7 consecutive days. Excisional biopsies of the tongue were performed and the extent of the lesion area, inflammatory infiltrate and tissue remodeling were assessed through histopathological analyzes, biochemical measurements of cytokines (TNF-α, TGF-β1, VEGF), NAG enzymes (N-acetylglucosaminidase) and MPO (Myeloperoxidase). Additionally, potencial beneficial effects on the oral microbiota were also evaluated.
Results: The application of topical L-Glutamine significantly reduced the area of the lesion and there was a change in the inflammatory profile, with an improvement in tissue remodeling in tongue lesions as well as the reestablishment of the resident oral microbiota.
Conclusion: L-glutamine potentiated tissue repair of oral mucositis induced in animals treated with 5-FU.
