Resumo: Objectives: Melanocytes, derived from the neural crest, synthesize melanin for skin photoprotection but also exist in sun-unexposed areas, where their roles are poorly understood. This study reviews melanocyte functions across the human body. Methodology: A literature review was conducted on PUBMED, focusing on articles discussing the functions of human melanocytes. Results: Melanocytes produce eumelanin (photoprotective), pheomelanin (phototoxic), and neuromelanin. Neuromelanin is produced in the brain, where it plays a role in neurodetoxification, neuroendocrine, and antioxidant functions. Elsewhere in the body, the balance between eumelanin and pheomelanin serves other functions. In the skin, melanocytes contribute to photoprotection, thermoregulation, protection against reactive oxygen species and free radicals, sequestration of metal ions, binding to drugs and organic molecules, and innate immune response. In the cochlea, they contribute to hearing and balance by participating in endolymph synthesis. In the heart, they are involved in the mechanical functioning of valves and atrial arrhythmia. In the eye, they act as optical absorbers. In the oral cavity, melanocytes can act in the immune-inflammatory response and have neuroendocrine, angiogenic, and antioxidant activities. Conclusions: Melanocytes perform multiple functions beyond photoprotection and are found in various tissues of the human body. Support: CAPES; FAPEMIG.
