Resumo: Objectives: Melanocytes produce melanin and are primarily found in the epidermis, hair follicle, and eye, where photoprotection is their primary function. Melanocytes can also be found in sun-protected sites such as oral mucosa, ears, meninges, heart, adipose tissue, gonads, lungs, and possibly bones, where the distribution and function remain poorly understood. This study investigates the expression of two melanogenic genes in six different sub-sites of the oral mucosa. Study Design: FFPE samples were selected including eight specimens from each sub-site of the oral cavity: tongue, buccal mucosa, hard palate, soft palate, floor of the mouth, and gingiva, comprising forty-eight cases in total. RT-qPCR was performed to detect gene expression of TRP2 and MiTF. Results: Overall, RT-qPCR showed low TRP2 and MiTF gene expression in all investigated sites of the oral cavity, and gingiva had higher TRP2 gene expression compared to the tongue (p<0.05). Conclusion: The low expression of TRP2 and MiTF genes is in accordance with the low melanin production by oral melanocytes. Support: CAPES; FAPEMIG.
