Título: A PROTOCOL FOR DYSPLASTIC AND MALIGNANT TRANSFORMATIONS IN NORMAL KERATINOCYTES, SIMULATING TOBACCO EFFECTS

Resumo: Objective: The aim of this study was to develop a protocol for inducing malignant and dysplastic transformations in normal keratinocytes in vitro using a specific concentration of 4-nitroquinoline (4-NQO) and to assess resultant morphological alterations. Study design: HaCaT cells, immortalized human keratinocytes, were selected for induction of changes using different concentrations of 4-NQO. Histological evaluation of cells was conducted using hematoxylin and eosin staining, and immunohistochemistry was performed with the Ki-67 marker. Results: Although in histological analysis, we can observe a reduction in the number of cells, the results indicate that a concentration of 1.3µM 4-NQO induced significant morphological changes indicative of dysplastic transformation, while 2.6µM resulted in significant histological alterations suggestive of malignancy. In immunohistochemical analysis for malignancy, not enough positive marking was obtained. Conclusion: In conclusion, the in vitro 4-NQO-induced dysplastic transformation protocol is feasible and can serve as a manageable model for further studies, also can be effective for an early diagnosis study since it serves as an indication for a pre-malignancy due to the observation of the dysplasia's degrees and even a malignancy.
