Resumo: Background: Epigenetic events play an important role in cancer behavior. Indeed, drugs that target epigenetic modifiers of histones presented effective inhibitory outcomes in many head and neck cancer. However, mucoepidermoid carcinoma (MEC), which is the most frequent salivary gland malignant tumor presented few effective targeted therapies. In this way, high-throughput screening of histone modifiers drugs was conducted to identify novel compounds able to manage MEC. Study design: MEC cells and their cancer stem cells (CSCs) were screened for changes in cell death. A high-precision liquid handler was used to seed the cells and administer drugs in a high throughput format. ALDH levels and clonogenic potential were also investigated. All data were evaluated using ImageXpress from molecular devices.Results: Four drugs were identified as a promisor therapeutic candidate, by the ability to disrupt MEC. Interestingly, the ALDH levels were significantly reduced just for drugs selected based on CSCs, while the clonogenic potential was reduced after the treatment with all selected drugs.Conclusion: Administration of histone modification drugs can effectively and individually target CSC and non-CSC from mucoepidermoid carcinomas.FAPESP 2021/13381-9, 2019/06597-5, 2016/0710-4
