Título: OSTEOPONTIN EXPRESSION IN ORAL POTENTIALLY MALIGNANT DISORDERS AND ORAL SQUAMOUS CELL CARCINOMA

Resumo: Objective: To assess the immunohistochemical expression of osteopontin (OPN/SPP1) in oral potentially malignant disorders (OPMD), as a potential predictor of malignant transformation to squamous cell carcinoma (SCC). Study design: 55 patients diagnosed with OPMD or SCC were selected, and divided into five groups based on its histological features: I - no dysplasia; II - mild/moderate dysplasia; III - severe dysplasia/carcinoma in situ; IV - microinvasive SCC; V - invasive SCC. Clinical data was retrieved, and histological and immunohistochemical analysis were performed. Results: Females represented 52.7% of the sample and mean age of the patients was 58 years (ranging from 24 to 90 years). Leukoplakias represented 60% of the sample, with tongue being the most affected site (29.1%). Histologically, 85.5% of the specimens presented epithelial dysplasia. OPN expression was inconsistent among groups I, II and III. Variable OPN expression was observed in all cases from groups IV and V, with expression predominantly localized in the middle third of the epithelium and in SCC tumor invasion front. Conclusion: The role of OPN in predicting the risk of malignant transformation remains inconclusive, but the preliminary results suggest its usefulness as an early marker for invasion.
