Resumo: To analyze protein immunoexpression and single-nucleotide variants (SNV) in a case series of odontogenic ghost cell lesions (OGCL).
Thirty-seven calcifying odontogenic cysts (COC) and three dentinogenic ghost cell tumors (DGCT) were selected from 65 cases of OGCL. Protein immunoexpression was evaluated using a broad panel of antibodies; SNV were studied in the ctnnb1 gene in 17 COC cases.
In both entities, COC and DGCT, the ameloblastic/ameloblastomatous epithelium and ghost cells stained positive for AE1-AE3 (40/40), amelogenin (40/40), beta-catenin (40/40), E-cadherin (40/40), S100 (22/40), vimentin (15/40), and CK19 (2/40). TOM20 (40/40), Bcl-2 (40/40), BRAF-V-600E (4/40), and p63 (1/40) were only positive in the ameloblastic/ameloblastomatous epithelium, and lysozyme (40/40) and CD68 (35/40) were positive in the ghost cells. CD1a was negative. Ki67 was ˂1% in COC and ≤5% in DGCT. No SNV were detected in ctnnb1, except for a possible SNV in codon 38.
The protein immunoexpression observed in ghost cells confirms an epithelial origin and suggest the possibility that these cells result from a degenerative process with an increase in lysosomes and accumulations of protein (keratins, amelogenins). In our case series, mutations in ctnnb1 do not seem to be involved in the etiopathogenic/tumorogenic process of these pathologies.
