Título: Organic depletion of salivary proteins after head and neck radiotherapy: preliminary results

Resumo: Objectives: To evaluate salivary protein quantification before, at the conclusion, and one year following head and neck radiotherapy (HNRT) for oral and oropharyngeal squamous cell carcinoma (OOSCC).Study design: Salivary flow rates and protein estimation using the Bradford assay were performed. 10 µg of total protein was used as standard for comparison. Statistical analysis encompassed the Shapiro-Wilk and Friedman tests followed by Wilcoxon tests with Bonferroni correction. Results: Significant decline in salivary flow at the end of RT (1.9 mL ± 1.3, p=0.011) and one year after RT (0.8 mL ± 0.9, p=0.002) compared to pre-HNRT (3.8 mL ± 2.2) and between the end of HNRT and one year after HNRT (p=0.001) was observed. Increased saliva required to obtain 10 µg of protein (p=0.001) was noted and supported by higher saliva volume required at the end of HNRT (47.6µL ± 42.2, p=0.003) and one year post-HNRT (63.2 µL ± 36.2, p=0.003) compared to pre-HNRT (15.7 µL ± 16.4).Conclusions: Non-stimulated salivary flow and protein content at the end and one-year post-HNRT were significantly impaired. Reduction in the total amount of salivary proteins may be regarded as an important factor to be considered in future studies regarding salivary proteomic analysis post-HNRT.
