Título: CURCUMIN TRIGGERS ROS-MEDIATED APOPTOSIS AND SUPPRESSES TUMOR GROWTH IN METASTATIC ORAL SQUAMOUS CELL CARCINOMA

Resumo: Objective: In this experimental study, we evaluated the in vitro and in vivo antineoplastic effects of curcumin in metastatic oral squamous cell carcinoma (OSCC) cells.
Study design: The cytotoxicity of curcumin was evaluated in 2D and 3D models, using the CellTiter-Glo® assay (Promega). Morphological parameters (FSC and SSC), apoptosis, and production of reactive oxygen species (ROS) were analyzed by flow cytometry. Furthermore, the morphological analysis of the spheroids was carried out using optical microscopy. In sequence, an analysis of antitumor efficacy in vivo was performed in a xenotransplant model using an OSCC cell line.
Results: Curcumin showed significant cytotoxic activity in both models, induced apoptosis, and increased ROS production. The curcumin-induced apoptosis was prevented by pretreatment with an antioxidant (N-acetyl-l-cysteine). Morphological analysis of the spheroids demonstrated cellular disaggregation, disorganization of the spheroid structure, and destruction of the proliferative zone's outermost layers. In vivo, the compound suppressed tumor growth, and tumor emboli and metastatic nodules were more evident in the lungs of the negative control group.
Conclusions: Curcumin showed promising cytotoxic activity by promoting ROS-mediated apoptosis and suppressing tumor growth in vivo.
