Título: IMPACTS OF ARTEMISININ AND CISPLATIN ON ORAL LEUKOPLAKIA: INSIGHTS FROM AN IN VITRO AND IN VIVO STUDY

Resumo: Objectives: To investigate the impact of artemisinin (ART) and cisplatin (CSP) on apoptosis and immunogenic cell death (ICD) in vitro, and to explore their potential in preventing oral leukoplakia (OL) progression in vivo. Study design: HaCat, DOK, and SCC-180 cell lines were exposed to ART, CSP, and ART+CSP to assess cytotoxicity, genotoxicity, cell migration, and the expression of proteins associated with apoptosis and ICD. Additionally, an in vivo experiment the OL was induced in 50 mice using 4NQO. These mice received treatment with ART and CSP, and microscopical analysis was performed. Results: In vitro, both CSP and CSP+ART showed dose-dependent cytotoxicity and decreased migration of SCC-180 cells. None of the treatments exhibited genotoxic effects. CSP decreased the expression of High-mobility group box-1 (HMGB1) protein in SCC-180 cells. In vivo, treatment with ART and CSP resulted in a delay in the progression of OL, but only CSP prevented progression to oral squamous cell carcinoma (OSCC). Conclusion: In vitro, CSP demonstrated a reduction in immunogenic pathways in SCC-180 cells while also inhibiting their migration. Despite significant side effects, CSP effectively prevented the malignant progression of OL in vivo.
