Título: DIGITAL ANALYSIS OF p53 IN TRAUMATIC REACTIVE LESIONS, LEUKOPLASIA AND ORAL CANCER

Resumo: Objective: The aim of this study was to analyze p53 immunostaining in Oral Potentially Malignant Disorders (OPMD), reactive fibrous hyperplasias (RFH), chronic traumatic ulcers (CTU), oral squamous cell carcinomas associated with OPMD (OSCC-seq), and those not associated with OPMD or de novo (OSCC-dn).
Study design: From 2016 to 2022, 20 patients from each of the following groups were included: OSCC-seq, OSCC-dn, CTU, Leukoplakia, and RFH. p53 immunohistochemistry was performed on biopsies, and digital image analysis was conducted using ImageJ Version 1.54f software to compare the percentage of epithelium with positive p53 staining (%p53+). The study met ethical standards.
Results: The %p53+ for OSCC-seq, OSCC-dn, CTU, Leukoplakia, and RFH were 2.21%, 1.67%, 0.84%, 0.59%, and 0.39%, respectively, with statistically significant differences (ANOVA, p=0.0162). According to the T Test, there were no differences between OSCC-dn and OSCC-seq (p=0.53), and between leukoplakia and CTU (p=0.56). Based on the tertile distribution of %p53+, CTU was more similar to OSCC-dn and OSCC-seq than leukoplakia and RFH. The suprabasal and parabasal location of p53 was similar between CTU and leukoplakia.
Conclusion: A subgroup of CTU presents p53 expression similar to leukoplakia and even to OSCC, which could reflect phenomena linked to early carcinogenesis.
