Resumo: Objectives: The present study aimed to establish two new cisplatin-resistant oral squamous cell carcinoma (OSCC) cell lines and investigate their biological characteristics.Study Design: OSCC cell lines SCC-9 and HSC-3 were exposed to cisplatin using stepwise dose escalation methods, and a total of two cisplatin-resistant sublines were generated, after determination of 50% inhibitory concentration (IC50). The cisplatin-resistant cells were subjected to morphological analysis, migration and invasion assays and evaluation of epithelial-mesenchymal transition (EMT)-related phenotypes.Results: The results demonstrated a significant increase in resistance to cisplatin-induced cell death in chemoresistant cell lines, which was accompanied by the acquisition of a fusiform shape and increased potential of migration and invasion. Furthermore, a significant increase in the expression of EMT markers and MMP-2 and -9 activities were observed.Conclusions: This study provides novel insights into OSCC cisplatin resistance, providing a valuable tool for investigating the molecular pathways and putative markers of EMT that may be associated with this resistance phenotype in oral cancer.Funding: FAPESP 2022/00994-5 and 2022/08908-0.
