Título: Effect of photobiomodulation on cytokine expression by HaCAT exposed to supernatant from PBMC stimulated with LPS.

Resumo: Objective: The aim of the study was to evaluate the effects of red wavelength laser (660nm), infrared (880nm) and the association of both on the modulation of inflammatory cytokines produced by HaCaT keratinocytes challenged by cytokines of human monocytes stimulated by LPS.
Study Design: human peripheral blood cells were collected, isolated and stimulated by LPS. The supernatant was used to stimulate HaCaT cells. HaCaT cells were exposed twice to diode laser (InGaAlP, 20mW, 0.71W/cm², 0.028cm²), at 5 J/cm² density, before challenge with cytokines. After four hours, the supernatant of the HaCaT cells was measured using CBA Multiplex assay. Cell viability was performed using MTT test.
Results: The energy density of 5J/cm² and stimulation with cytokines did not affect the cell viability. The infrared laser increased the production of IFN-α2, IFN-γ, TNF-α, MCP-1, IL-6, IL-8, IL-10, IL-12p70, IL-17A, IL-23 and IL-33 (P<0.05) in relation to the control group. Red laser on HaCaT cells increased the expression of IFN-α2 cytokine compared to control (P<0.05).
Conclusion: The red, infrared and the two lasers simultaneously applied at an energy density of 5 J/cm² seem to have the ability to modulate inflammation on HaCaT cells challenged by human monocyte cytokines.
