Título: Different microRNA expression profiles associated with transformation of conventional and proliferative verrucous leukop

Resumo: Objective: Characterize the microRNA expression profile in conventional oral leukoplakia (COL) and proliferative verrucous leukoplakia (PVL), with and without malignant transformation (MT), and its changes in relation to malignancy.
Methods: A multicenter case-control study of the microRNA expression profile in 62 samples: 26 COL (16 with MT and 10 without MT), 30 PVL (15 with MT and 15 without MT), and 6 controls (C). The expression of 768 mature microRNAs was analyzed using low-density TaqMan arrays.
Results: COL without MT showed no significant differences in microRNA profile compared to the control. COL with MT showed 6 overexpressed microRNAs compared to the control, and 85 differentially expressed microRNAs compared to the COL without MT. PVL showed extensive microRNA dysregulation compared to the control, with 152 differentially expressed microRNAs in PVL without MT and 234 in PVL with MT. Direct comparison between COL and PVL without MT showed 216 differentially expressed microRNAs.
Conclusions: COL and PVL exhibit distinct microRNA expression profiles. COL acquires molecular alterations during its MT, while PVL presents an early altered profile of microRNAs related to oral carcinogenesis, demonstrating that these are two molecularly distinct disorders despite their clinicopathological appearance.
