Título: REGULATION OF METABOLIC GENE EXPRESSION BY PERIODONTAL PATHOGENS IN HEAD AND NECK SQUAMOUS CELL CARCINOMA

Resumo: Objectives: Recent evidence highlights metabolic reprogramming as a key driver of cancer progression. This study aimed to investigate whether Fusobacterium nucleatum (F. nucleatum) and Porphyromonas gingivalis (P. gingivalis) modulate the expression of metabolic genes in head and neck squamous cell carcinoma (HNSCC) cells.

Study Design: FaDu squamous cell carcinoma cells were exposed to F. nucleatum and P. gingivalis at a multiplicity of infection of 100. The cells were harvested at 6 and 24 hours. Total RNA was extracted, and expression of metabolism-related genes, including SMS, ALDH2, KYNU, ACAT1, and PLD2, was evaluated using semiquantitative RT-PCR.

Results: Both pathogens caused time-dependent changes in gene expression. At 6 hours, polyamine (SMS), tryptophan (KYNU), and lipid signaling (PLD2) genes were significantly downregulated, especially by P. gingivalis. At 24 hours, genes involved in polyamine and tryptophan metabolism (SMS, KYNU) and lipid/aldehyde metabolism (ACAT1, ALDH2) were upregulated by both bacteria, with stronger induction by F. nucleatum. PLD2 stayed suppressed with P. gingivalis but increased with F. nucleatum at 24 hours.

Conclusions: These preliminary findings suggest that periodontal pathogens differentially modulate metabolic gene expression in a time-dependent and pathogen-specific manner. Further studies are required to elucidate the underlying mechanisms and their relevance to HNSCC.
