Título: IN VITRO ANTITUMOR EFFECTS OF BRAZILIN ON CANCER STEM CELL PHENOTYPE IN ORAL SQUAMOUS CELL CARCINOMA

Resumo: This study aimed to evaluate the in vitro effects of the isoflavonoid Brazilin (BRA), alone and in combination with Cisplatin (CIS), on tumor behavior and cancer stem cell (CSC) phenotype in oral squamous cell carcinoma (OSCC) cell lines. SCC-9 (primary tumor) and LN-1A (metastatic) cells were treated with IC50 doses of BRA and CIS. Migration (wound healing), invasion (Matrigel®-coated transwell), clonogenicity, and self-renewal (tumorsphere formation) were assessed. The expression of stemness markers (CD44, BMI1, NANOG, OCT4) was analyzed by RT-qPCR, and the CD44/ESA profile was evaluated by flow cytometry. Brasilin reduced cell growth, migration, invasion, and the formation of colonies and tumorspheres in both cell lines, and shifted the CD44/ESA profile to a less invasive phenotype. The combination of BRA and CIS enhanced the observed antitumor effects, including a slight reduction in OCT4 expression in LN-1A cells. In conclusion, Brazilin exhibits significant antitumor activity in OSCC, impacting tumor progression and cancer stem cell-related features. Its combination with Cisplatin further potentiates these effects, suggesting a promising therapeutic approach.
