Título: EVALUATION OF HISTOPATHOLOGICAL PATTERNS IN ADVANCED OSCC AND CHARACTERIZATION OF CAFs IN VITRO

Resumo: Objective: To evaluate histopathological patterns in cases of advanced OSCC, refractory and treatment-responsive. Concomitantly, to characterize CAFs in vitro.
Study design: A translational analytical observational study combined with an in vitro experimental stage.
Results: The analysis of depth of invasion (DOI) ranged from 0.35 to 4.3 mm for responsive cases and from 1 to 3.1 mm for refractory cases, showing no relationship between higher DOI and treatment refractoriness. In contrast, the evaluation of extranodal extension (ENE) revealed that most responsive patients did not present ENE, while refractory cases demonstrated the presence of ENE, suggesting an association with poorer therapeutic response in these patients. In the experimental stage, the characterization of CAFs by flow cytometry showed expression of α-SMA in two of the three passages analyzed, which were selected for subsequent studies.
Conclusion: Therefore, DOI did not differentiate responsive and refractory cases, while the presence of ENE showed a trend towars of association with refractoriness. The caracterization of fibroblasts as CAFs is a valuable model for studing the tumor mictoenvitonment in vitro. These findings reinforce the relevance of integrating histopathological and experimental data in OSCC. Project supported by the São Paulo Research Foundation - FAPESP (#2024/20525-5).
