Resumo: Objectives: To develop a decellularized human-based extracellular matrix (dhECM) hydrogel derived from normal human tongue mucosa and evaluate its ability to support physiologically relevant 3D in vitro modeling of oral squamous cell carcinoma (OSCC). Methods: Normal tongue mucosa was obtained from donor patients during autopsy (n=6), subjected to decellularization, and processed to generate the dhECM hydrogel. Organotypic 3D models were established by populating the hydrogel with normal oral fibroblasts (NOFs) and OSCC cell lines. Results: DNA quantification confirmed successful decellularization, with no detectable residual DNA. Histological analyses using hematoxylin and eosin, Alcian Blue, Sirius Red, and Masson's Trichrome demonstrated preservation of major extracellular matrix components, including collagen and proteoglycans, within the decellularized scaffold. Organotypic OSCC models generated using dhECM exhibited epithelial growth, organization, and invasion patterns closely resembling native tumor histomorphology. Conclusion: The human tongue-derived dhECM represents a biologically relevant scaffold that preserves essential extracellular matrix components and supports in vitro organotypic OSCC modelling with in vivo-like histological characteristics. This platform may provide a valuable tool for investigating tumor-stroma interactions and for the preclinical study of oral cancer biology.
