Título: E6/E7 mRNA IN SITU HYBRIDIZATION FOR DETECTION OF HIGH-RISK HPV IN OROPHARYNGEAL SQUAMOUS CELL CARCINOMA

Resumo: Objective: This study aimed to evaluate the application of in situ hybridization using E6/E7 mRNA probes to identify the frequency of high-risk HPV (HR-HPV) transcriptionally active and its use as a prognostic biomarker in oropharyngeal squamous cell carcinoma (OPSCC).
Study design: Thirty-six OPSCC samples were used to construct a tissue microarray, and the slides were submitted to an in situ hybridization detection method for HPV E6/E7 mRNA and compared to p16 expression by immunohistochemistry. HPV status was designated using binary classification. Multiple logistic regression was used to analyse the association of HPV with clinical features and other risk factors. A significance level of p ≤ 0.05 was considered.
Results: HPV frequency in oropharyngeal squamous cell carcinoma was 11,1%. There was no association between HR-HPV, clinical variables, and known risk factors. The mean overall survival (OS) was 22.5 months for patients with HPV-negative tumors and 39.5 months for those with HPV-positive tumors (p=0.340), and the mean disease-free survival (DFS) was 44.4 months for HPV-negative patients and 24.4 months for HPV-positive patients (p=0.810)
Conclusions: A low frequency of E6/E7 mRNA was found in OPSCC, which supports the evidence that HPV-driven cancer of the oropharynx is uncommon in Brazil. FAPES and HORIZON2020.
