Título: PROTEOMIC PROFILING OF SALIVARY GLAND CARCINOMAS REVEALS PUTATIVE SUBTYPE-RESTRICTED PROTEINS

Resumo: Objective: To identify proteins exclusively expressed across subtypes of salivary gland carcinomas and characterize subtype-restricted proteomic patterns. Study Design: A proteomic analysis using mass spectrometry with label-free quantification was performed on 25 tumors across five histological groups: myoepithelial carcinoma, adenoid cystic carcinoma, epithelial-myoepithelial carcinoma, salivary duct carcinoma, and adenocarcinoma, not otherwise specified. Data was processed using MaxQuant and aligned to UniProt. Reverse identifications and entries based solely on modification sites were excluded. Proteins were filtered to retain at least one valid value per group, and contaminants were manually removed. Results: Across the five groups, subtype-restricted protein detection patterns were identified, with the most prominent findings in specific subtypes. In myoepithelial carcinomas, NPM3 and PYCRL were consistently detected; PYCR proteins are linked to proline metabolism and have been associated with poor prognosis. In salivary duct carcinomas, PAPSS2, TFRC, and REEP6 showed homogeneous detection. PAPSS2 has been implicated in invasion and migration, TFRC in hypoxia-related pathways, and REEP6 in tumor aggressiveness. Conclusion: Distinct subtype-restricted proteomic patterns were identified, including proteins not previously reported in this context, reinforcing their relevance as targets for further validation and subtype-oriented investigation.
