Resumo: Objective: To investigate the pattern of immunoexpression for β-catenin, CK-19, and Bcl-2, correlating it with clinical and histopathological features of calcifying odontogenic cysts. Study design: Twenty COCs were analyzed for the immunoreactivity of β-catenin, cytokeratin-19 (CK-19), and Bcl-2. The evaluation considered the percentages of cytoplasmic and nuclear immunoexpression in epithelial and ghost cells. Results: Weak β-catenin immunoreactivity was observed in the basal and suprabasal layers of the lining epithelium in most cases. In contrast, intense immunoreactivity was observed in ghost cells. CK-19 showed strong expression in basal and suprabasal cells. In ghost cells, low staining was observed. Bcl-2 demonstrated strong expression in basal cells, with a variable pattern in suprabasal cells, ranging from weak to strong to absent, and was not detected in ghost cells. Conclusion: The findings suggest the involvement of β-catenin in the regulation of cell proliferation and differentiation processes in COC. The expression of CK-19 reinforces the presence of odontogenic epithelial differentiation, possibly related to an ameloblastic phenotype. Bcl-2 appears to be involved in maintaining cell viability in the epithelial proliferative compartment. Taken together, the results corroborate the current understanding of the World Health Organization, indicating a predominantly non-neoplastic behavior for this lesion.
