Título: COMPARATIVE ANALYSIS OF APOPTOSIS-ASSOCIATED GENE EXPRESSION IN ODONTOGENIC KERATOCYST AFTER MARSUPIALIZATION

Resumo: OBJECTIVE: To compare the expression of 96 apoptosis-related genes in OKC epithelium after marsupialization followed by validation using fluorescent immunohistochemistry (FIHC). STUDY DESIGN: OKC samples were analyzed before and after marsupialization. RNA was extracted for RT-qPCR using Human Apoptosis 96-Well Plate TaqManT Array (Thermo Fisher Scientific). Data were normalized and analyzed in R. Differential expression was assessed between groups; genes with p < 0.01 and log₂FC < −3 were selected for further analysis, including FIHC. RESULTS: Differential expression analysis identified seven BCAP3, BCL2, BIRC2, CRADD, HIP1, ICEBERG, TNFRSF25 significantly downregulated genes in mOKC. Top differentially expressed genes (BIRC2, HIP1, BCL2, and Caspase-3) were validated by FIHC. Results showed a significant increase in Caspase 3 expression while BCL2 expression decreased after marsupialization. CONCLUSION: Marsupialization promotes apoptosis via Caspase 3, downstream of p53 and BCL2, providing a new insight into the mechanisms of OKC regression. Funding: FAPEMIG and CNPq.
