Resumo: Objective
To investigate migratory behavior and the presence of amoeboid features in cisplatin-resistant oral squamous cell carcinoma (OSCC) cell lines.

Study Design
In vitro experimental study using cisplatin-sensitive and cisplatin-resistant OSCC cell lines.

Results
Induction of chemoresistance increased cisplatin IC50 in resistant SCC-9 and HSC-3 cell lines compared with parental cells. Resistant cells exhibited morphological changes consistent with epithelial-mesenchymal transition (EMT), including elongated and spindle-shaped morphology. Gene expression analysis confirmed the EMT profile, showing increased VIMENTIN and TWIST expression and reduced E-cadherin expression in resistant cells. In addition, rounded cells displaying membrane "blebs" were observed, suggesting acquisition of an amoeboid phenotype. Analysis of amoeboid-related markers (GENE 1, 2, and 3) demonstrated a significant increase in GENE 1 transcript in resistant SCC-9 cells and a trend toward increased GENE 2 transcript in the same lineage. Quantitative cell migration assays showed no statistically significant differences between resistant and parental cells, although resistant cells showed a consistent trend toward faster wound closure.

Conclusion
Cisplatin resistance in OSCC is associated with migratory plasticity, including EMT characteristics and a possible transition toward an amoeboid phenotype, suggesting that adaptation to chemotherapy may favor alternative mechanisms of tumor migration and invasion.
