Título: ANALYSIS OF PYROPTOSIS AND NECROPTOSIS INDUCED BY DYNAMIC THERAPIES IN ORAL SQUAMOUS CELL CARCINOMA: AN IN VITRO STUDY

Resumo: Objective
To verify and quantify the regulated cell death (RCD) pyroptosis and necroptosis induced by 5-ALA-mediated photodynamic therapy (PDT), sonodynamic therapy (SDT) and sonophotodynamic therapy (SPDT) in an oral squamous cell carcinoma (OSCC) cell line.

Study Design
SCC9 cells were treated with 5-ALA-mediated PDT, SDT, or their combination (SPDT) for 1, 3, or 5 minutes. Cell viability was assessed by MTS assay, membrane integrity via lactate dehydrogenase (LDH) release, and GSDMD and pMLKL expression via immunofluorescence. All treatments were compared to untreated controls.

Results
Exposure to LED or ultrasound in the absence of 5-ALA did not reduce cell viability. In contrast, PDT, SDT, and especially SPDT treatments led to dose-dependent cytotoxicity. SPDT significantly reduced viability compared to PDT or SDT alone, with marked morphological features suggestive of RCD. LDH release was significantly elevated in all treatment groups versus control, with SPDT inducing the highest levels. Immunofluorescence analysis revealed significantly increased expression of GSDMD and pMLKL SCC9 cells post-treatment, particularly in the SPDT group.

Conclusion
These findings support SPDT as a promising therapeutic strategy for OSCC, enhancing the occurrence of pyroptosis and necroptosis compared to PDT and SDT alone. Targeting RCD mechanisms is advantageous due to increased immunogenicity.
