Título: NANOENCAPSULATED CINNAMALDEHYDE AS AN ADJUVANT THERAPY FOR CHEMICALLY INDUCED ORAL SQUAMOUS CELL CARCINOMA OF THE TONGUE

Resumo: Objective: To evaluate the effect of polymeric nanocapsules containing cinnamaldehyde on chemically induced tongue oral squamous cell carcinoma in mice.

Study design: Experimental study involving development and physicochemical characterization of cinnamaldehyde-loaded polymeric nanocapsules. In vitro assays were performed using SCC9 oral squamous cell carcinoma cells and HaCaT keratinocytes to assess cytotoxicity, proliferation, and migration. In vivo analysis used a murine model of chemically induced tongue oral squamous cell carcinoma. Histopathological examination was performed with hematoxylin and eosin staining. Bioinformatics, molecular docking, and RT-qPCR were used to investigate molecular mechanisms associated with tumor progression.

Results: The nanocapsules showed a mean diameter of 257.2 nm. NC-CNM at 50 μg/mL exhibited cytotoxic activity against SCC9 cells, reducing proliferation and migratory capacity in vitro. In animals, NC-CNM at 50 mg/kg inhibited tumor invasion in the tongue and reduced MMP2 expression, a marker associated with extracellular matrix degradation and invasion, while also reinforcing its potential to modulate pathways related to tumor aggressiveness.

Conclusion: Nanoencapsulated cinnamaldehyde demonstrated promising antitumor activity and may represent an innovative adjuvant strategy for tongue oral squamous cell carcinoma treatment.
