Título: Melatonin-Copper Composites and CEMP1 as Biomarker of Tumor Plasticity: Preliminary Antitumor Evidence

Resumo: Cervical cancer remains a major global health challenge, requiring more selective therapeutic strategies beyond platinum-based treatments. Melatonin-copper composites show promising antitumor activity by targeting tumor-specific vulnerabilities in heterogeneous models, including MDA-MB-231 and MCF-7. CEMP1 emerges as a key regulator of proliferative and mesenchymal phenotypes, linked to the MAPK/ERK and Wnt/β-catenin signaling pathways, supporting a precision oncology approach. Cytotoxic effects of melatonin-derived carbon dot-copper(II) composites (m34 and m35) were evaluated in HeLa, MDA-MB-231, and MCF-7 cells using the MTT assay. Human dermal fibroblasts served as non-tumoral controls. Cells were exposed to 0.0625-1.0 mM concentrations, and IC₅₀ values were calculated. Statistical analyses included ANOVA, Bonferroni post-hoc tests, and effect size estimation. Both composites showed significantly higher cytotoxicity in tumor cells versus controls. In HeLa cells, m35 showed the greatest potency (IC₅₀ = 0.2035 ± 0.019 mM), outperforming m34 and CuCl₂, while melatonin alone was ineffective. Similar responses were observed in breast cancer models, with preserved HDF viability. Synergy analysis (δ = 0.425) supported supradditive effects. CEMP1 expression correlated with proliferative and mesenchymal traits. These composites exhibit potent, selective antitumor activity. CEMP1 may serve as a biomarker of tumor plasticity and aggressiveness, supporting further preclinical development of m35.
