Resumo: Objective: To analyze oncogene expression profiles in tobacco-associated oral squamous cell carcinoma (OSCC), comparing them with OSCC lesions potentially related to chronic mechanical irritation (CMI).
Study Design: A cross-sectional observational study was conducted through the retrospective selection of 40 formalin-fixed, paraffin-embedded (FFPE) tissue samples diagnosed as OSCC, comprising 22 tobacco-associated cases and 18 cases potentially related to chronic mechanical irritation. All samples were analyzed using the TaqMan® OpenArray® Oncogenes & Tumor Suppressor Genes panel on the QuantStudioT 12K Flex platform to assess the expression of 112 oncogenes.
Results: Following statistical (t-test) and bioinformatic analyses, the volcano plot revealed no statistically significant differences in gene expression levels (p > 0.05) between the groups (tobacco vs. CMI). Pearson's correlation analysis (r > 0.7) demonstrated a strong concordance between the groups (r = 0.82), consistent with the global gene expression profile observed in the heatmap. Hierarchical clustering using the complete linkage method did not identify distinct clusters between the groups.
Conclusion: The findings suggest a high degree of similarity in the gene expression profiles of the two groups analyzed in this study.
