Resumo: Objective: To evaluate oral microbiological composition and association with risk factors, aiming to identify microbial profiles as potential biomarkers of tumor risk and progression.Methods: Descriptive observational case-control study included 47 patients: control (reactive hyperplastic lesions, n = 24), potentially malignant disorders (OPMD, n = 17), squamous cell carcinoma (SCC, n = 6). Samples from healthy mucosa, lesions, and saliva were obtained. Presence of Candida spp., Lactobacillus spp., Staphylococcus aureus, Staphylococcus spp., Escherichia coli, and Enterobacteriaceae was assessed using selective culture media. Tobacco, alcohol, and yerba mate consumption were recorded. Fisher's exact test and odds ratios (OR, 95% CI) were calculated. HPV-PCR was performed in 34 samples.Results: Significant associations were found between sample type and microbial frequency. Candida spp., Staphylococcus aureus, and Staphylococcus spp. were more frequently detected in saliva (p < 0.05). Tobacco use was associated with higher S. aureus detection (OR = 3.39; p = 0.004). Alcohol consumption increased S. aureus detection (OR = 9.53; p < 0.001) and decreased Staphylococcus spp. (OR = 0.25; p < 0.001). Yerba mate showed no association. HPV-PCR negative in all samples.Conclusion:Oral microbiota varies by sample type and is influenced by tobacco and alcohol, supporting its potential as a biomarker of carcinogenesis.
