Título: LONG NON-CODING RNAs AS POTENTIAL BIOMARKERS OF CHEMORESISTANCE IN ORAL CARCINOMA

Resumo: This work investigated the role of long non-coding RNAs in cisplatin resistance of oral carcinoma. RNA-seq investigated differences in the profile of lncRNAs between wild-type (WT) and cisplatin-resistant (CisR) CAL27 and SCC9 cell lines, and quantitative PCR was used to validate lncRNAs differentially expressed. In the CAL27 CisR cell line, 50 elevated lncRNAs were identified associated with chemoresistance and cancer stem cell accumulation, in addition to 48 decreased lncRNAs. In the SCC9 CisR cell line, 77 decreased, and 112 elevated lncRNAs were observed; among the elevated lncRNAs, 12 were associated with epithelial-mesenchymal transition, stem cell accumulation, and chemoresistance. Interestingly, LINC00973, LINC01133, and FGD5-AS1 were highly overexpressed in extracellular vesicles of both chemoresistant cell lines. Bioinformatic analysis of the correlation between LINC00973 and mRNAs indicated a positive correlation and direct interaction between LINC00973 and genes associated with the phosphatidylinositol-mediated signaling, and a negative correlation and direct interaction between LINC01133 and genes associated with the regulation of the circadian rhythm. Cisplatin resistance may be associated with alterations in the expression of lncRNAs, regulating mRNAs involved in vital metabolic processes. Moreover, as lncRNAs can be highly expressed in extracellular vesicles, they could work as non-invasive biomarkers for predicting chemoresistance in oral carcinoma.
