Título: LACK OF ERα IMMUNOEXPRESSION SUGGESTS OESTROGEN-INDEPENDENT MECHANISMS IN THE REGRESSION OF CHERUBISM AND BROWN TUMOURS

Resumo: Objective: Cherubism-associated giant cell lesions and non-ossifying fibroma are benign bone lesions with overlapping histological features and tendency for spontaneous regression after puberty. Oestrogen plays a key role in bone homeostasis, and oestrogen receptor alpha (ERα) immunoexpression in non-ossifying fibroma suggests a role for oestrogen-mediated regulation in its regression. This study aimed to evaluate whether cherubism-associated giant cell lesions show ERα immunohistochemical positivity. Given their histological similarities and shared regression potential, brown tumours were also analysed. Study design: In addition to cherubism (n=8 samples, 6 patients) and brown tumours (n=7), central (n=9) and peripheral (n=7) giant cell granulomas of the jaws were included for comparison. All samples underwent immunohistochemistry using a rabbit monoclonal anti-ERα antibody (clone EP1) and nuclear staining was evaluated. Results: None of the samples of any of the studied entities demonstrated nuclear ERα immunopositivity. Conclusion: These findings indicate that regression in cherubism-associated giant cell lesions and brown tumours (upon PTH normalization) is unlikely to be mediated by ERα-dependent mechanisms.
