Título: LONG NON-CODING RNA MSC-AS1 ACTS AS AN ONCOGENIC DRIVER OF CHEMORESISTANCE AND TUMOR PROGRESSION IN ORAL CANCER

Resumo: Long non-coding RNAs are associated with cancer development, acting either as oncogenes or tumor suppressors. The long non-coding RNA Musculin Antisense 1 (MSC-AS1) confers chemoresistance in various tumor types. This project sought to identify the role of MSC-AS1 in oral carcinogenesis and its association with chemoresistance. NGS identified MSC-AS1 as overexpressed in chemoresistant cell lines. CRISPR-Cas9 was used to deplete MSC-AS1. Quantitative RT-PCR assays were performed to quantify MSC-AS1 in cells and within extracellular vesicles and genes associated with EMT. Immunofluorescence identified proteins associated with EMT. The spheroid assay investigated stem cell accumulation. Cell migration and invasion assay to assess the aggressiveness and malignancy of cells with and without MSC-AS1. Cell viability assay evaluated chemoresistance. Our results demonstrated that MSC-AS1 depletion resulted in reduced tumor stem cells, reduced migratory and invasive capacity, reversal of chemoresistance, and diminished transport of MSC-AS1 by extracellular vesicles. It is concluded that MSC-AS1 acts as oncogene and its depletion reversed the phenotype of tumor cells to a less aggressive state, proving to be a promising target in the treatment of this disease.
