Resumo: Objective: To investigate the effects of anastrozole (ANZ), a non-hormonal aromatase inhibitor, on morphological structure of major salivary glands (MSG) in a mice model. Study Design: Twenty 8 weeks-old male *C57/Bl6* mice were randomly allocated in groups ANZ- and vehicle-treated. ANZ groups received subcutaneous injections of ANZ at 0.3, 1.0 and 3.0 mg/Kg and were euthanized 30 days after. Glands were histologically processed and morphometrically analyzed for acini and duct counts, glandular area, acinar and ductal diameter and density. Results: Administration of ANZ in mice resulted in significant parenchymal atrophy within the sublingual and parotid glands, evidenced by decreased numbers of acini and ducts as well as a reduction in overall glandular area. Similar effects were observed at three different doses. The submandibular gland showed no significant morphometric changes. Conclusion: Results suggest detrimental effects of ANZ on MSG, underscoring the importance of clinical monitoring patients receiving medications that reduce the estrogen levels.
