Resumo: Objective: To evaluate the potential of keratinocyte and fibroblast spheroids associated with 3D bioprinting as a strategy for oral tissue regeneration after pathological conditions and surgical interventions. Study design: Keratinocytes (HaCaT) and fibroblasts (BJ) were used to generate monocellular and cocultured spheroids (1:1). After 4 days, spheroids were incorporated into a collagen-based bioink (Cultrex®) and bioprinted by extrusion. Morphology, viability, phenotypic markers, and cellular behavior before and after printing were assessed by immunofluorescence. Results: Spheroids showed viability, three-dimensional organization, and maintenance of epithelial and mesenchymal characteristics. After bioprinting, active cell migration, organized cellular areas, and early tissue reorganization were observed. Cocultured spheroids showed more coordinated behavior and greater structural integration than separately formed spheroids. Conclusion: Cocultured spheroid bioprinting is a promising approach for oral tissue regeneration and functional tissue reconstruction. Funding: NIH-NIGMS, CAPES, FAPESP.
