Título: EFFECTS OF CURCUMIN AND PHOTOBIOMODULATION THERAPY ON IRRADIATED OSTEOBLAST-LIKE CELLS: A PRELIMINARY ANALYSIS

Resumo: Objective: In vitro evaluation of the effects of curcumin and photobiomodulation therapy (PBMT) on irradiated immortalized osteoblast-like human cells (SaOS-2).
Study Design: SaOS-2 cells were exposed to a single dose of ionizing radiation (16 Gray) and treated with vehicle (control), curcumin and PBMT (660 nm; 30mW; 2.09, 2.79, 4.19, and 4.88 J/cm² corresponding to irradiation times of 3, 4, 6, and 7s, respectively). After 24 hours, MTT, nitric oxide (NO), and scratch assays were performed. Data were analyzed using ANOVA and Kruskal-Wallis tests in GraphPad Prism.
Results: Curcumin at 2.5 µM and PBMT at 3, 6 and 7s showed a trend toward increased mitochondrial activity. A significant increase in NO production was also observed with 5 µM (p<0.05), while a decrease was observed at 6s (p<0.05). Additionally, 2.5 µM curcumin and PBMT at 3 and 7s enhance wound closure compared to control, without statistical significance.
Conclusion: Tested doses of curcumin and PBMT did not significantly enhance mitochondrial activity after irradiation. However, some doses showed a non-significant trend toward increased wound closure, suggesting a limited but potential effect. Future research should further investigate these therapies in irradiated osteoblasts by exploring different cell lines and optimizing culture
conditions.
