Resumo: Objective: This study aimed to investigate the role of miR-134-5p in a mucoepidermoid carcinoma cell line to understand its molecular mechanisms and contribute to treatment stratification based on tumor aggressiveness. Study design: A previous study from our group identified miRNA expression profiles distinguishing non-neoplastic from tumor samples and metastatic from non-metastatic cases, leading to the selection of miR-134-5p for functional analysis. UM-HMC-3A cells were transfected with miRNA mimics and inhibitors, followed by validation of transfection by RT-qPCR and in vitro assays to assess proliferation, clonogenicity, migration, and invasion potential. Results: Transfection efficiency was confirmed by RT-qPCR, showing increased miR-134-5p expression in the mimic group and markedly decreased or undetectable expression in the inhibitor group. Data were normalized and analyzed by the 2⁻ΔΔCt method. Overexpression of miR-134-5p using the mimic reduced cell viability in the MTS assay and decreased clonogenic capacity. Additionally, reduced migratory and invasive potential was observed in Transwell assays. In contrast, inhibition of miR-134-5p led to increased proliferation, clonogenicity, migration, and invasion. Conclusion: We demonstrated that miR-134-5p acts as a tumor suppressor in mucoepidermoid carcinoma cells. These findings suggest its potential as a biomarker of tumor aggressiveness and a possible therapeutic target.
