Título: CYCLOPHOSPHAMIDE-INDUCED SALIVARY GLAND TOXICITY IN AN EXPERIMENTAL MODEL

Resumo: Objective: To investigate cyclophosphamide (CTX)-induced morphological alterations in the sublingual, submandibular, and parotid glands of BALB/c mice.
Study Design: Twenty 8 weeks-old male and female mice were randomly allocated in groups CTX- and vehicle-treated. CTX groups received 2 intraperitoneal injections of 200mg/kg (day 0) and 150 mg/kg (day 9) and were euthanized on day 14. Peripheral blood was collected for lymphocytes counting. Glands were histologically processed and morphometrically analyzed for acini and duct counts, glandular area, acinar and ductal diameter and density.
Results: The CTX-induced immunosuppression was confirmed by significant lymphocytopenia. Female CTX group exhibited marked parenchymal atrophy in the sublingual gland, drop in acinar number (~60%), duct count (~50%) and glandular area (~54%). In male CTX group, the submandibular gland showed a decreased duct-to-area ratio. The parotid gland showed no significant alterations in either sex.
Conclusion: Structural changes in salivary glands may lead to oral issues such as dry mouth and mucosal disruption in individuals treated with cyclophosphamide. The results emphasize the impact of biological sex and gland-specific responses in chemotherapy-related toxicity. Financial support: CAPES, CNPq, FAPEMIG
