Título: MICRORNA PROFILE ASSOCIATED WITH NEUROPATHIC PAIN IN PATIENTS WITH ORAL SQUAMOUS CELL CARCINOMA: A PCR ARRAY ANALYSIS

Resumo: The study aimed to evaluate the expression of microRNAs (miRNAs) associated with neuropathic pain and inflammation in individuals with Oral Squamous Cell Carcinoma (OSCC) using a PCR array. Formalin-Fixed Paraffin-Embedded (FFPE) samples from 8 individuals (5 with OSCC and 3 with healthy oral mucosa) were analyzed by quantitative PCR (qPCR) using a panel of 84 microRNAs related to neuropathic pain and inflammation. Expression analysis identified 54 overexpressed and 27 underexpressed miRNAs in OSCC compared with controls, with miR-451a, miR-31-5p, and miR-223-3p being the most highly overexpressed. Hierarchical clustering revealed distinct miRNA expression patterns, suggesting possible functional co-regulation. Functional analysis performed using an in silico Gene Ontology approach indicated that the most overexpressed miRNAs are involved in neuronal modulation and molecular pathways related to nociception and the opioid system, including the nociceptin receptor (OPR1). These findings suggest that a subset of inflammation- and pain-related miRNAs is differentially expressed in OSCC, supporting their potential role in the pathophysiology of cancer-associated pain. Furthermore, these miRNAs may represent promising candidates for prognostic biomarkers and for the development of adjuvant therapeutic strategies in cancer pain management.
