Título: THE IMPACT OF HDAC11 ON THE IFN-ALPHA PATHWAY IN CHEMORESISTANT ORAL SQUAMOUS CELL CARCINOMA

Resumo: To investigate the role of HDAC11 in oral squamous cell carcinoma (OSCC) cell lines resistant to cisplatin, focusing on the modulation of the IFN-alpha pathway and PD-L1-mediated signaling, and to determine the effects of genetic or pharmacological silencing on this pathway. OSCC cell lines CAL27 and SCC-9 (WT) were used, including cisplatin-resistant variants (CisR) and HDAC11-silenced cells (siHDAC11). In vitro experiments included siRNA-HDAC11 transfection and SAHA treatment in CisR cells, followed by gene and protein expression analyses and immunofluorescence. In vivo, CAL27 WT and CisR xenografts were established in BALB/c nude mice, and protein expression of HDAC11, IFN-alpha, IFITM1, and PD-L1 was evaluated by immunohistochemistry. HDAC11 was overexpressed in CisR cells. siRNA-HDAC11 reduced HDAC11, IFN-alpha, and IFITM1 levels, while SAHA also decreased PD-L1 expression. In vivo, CisR xenografts showed increased HDAC11 and IFN-alpha expression, which was reversed by SAHA treatment. No direct regulation of PD-L1 by HDAC11 was observed. HDAC11 may contribute to cisplatin resistance in OSCC by modulating IFN-alpha-associated pathways. Both siHDAC11 and SAHA demonstrated therapeutic potential by reducing chemoresistance markers and modulating the tumor microenvironment, suggesting a promising strategy for OSCC treatment.
