Resumo: Objective
Oral potentially malignant disorders (OPMDs) exhibit complex molecular alterations, involving microRNA dysregulation and changes in key proteins associated with cell cycle control, inflammation, and early carcinogenesis. The aim of this study was to evaluate the expression levels of selected microRNAs (miR‑21, miR‑31 and miR‑203) and proteins associated with oral carcinogenesis in normal oral mucosa (NOM), oral lichen planus (OLP) and oral dysplasia (OD).
Study design
Expression of miR‑21, miR‑31 and miR‑203 was analyzed by qPCR, and protein expression of p21, p53, p16, PTEN and HIF‑1α was assessed by immunohistochemistry. Comparisons among NOM, OLP and OD were performed using Kruskal-Wallis tests with multiple comparisons.
Results
OLP and OD showed significant overexpression of miR‑21 and miR‑31 compared to NOM, while miR‑203 expression was significantly reduced in both OLP and OD. Immunohistochemistry revealed increased expression of p21, p53, p16 and HIF‑1α in OLP and OD, whereas PTEN expression was preserved across groups. Integrated analyses demonstrated concordant molecular patterns consistent with progressive epithelial stress, metabolic activation and cell cycle dysregulation.
Conclusion
Distinct and biologically coherent miRNA and protein expression patterns characterize oral lichen planus and oral dysplasia, supporting a molecular continuum from chronic inflammation to early oral carcinogenesis.
