Título: INVESTIGATION OF MICRORNA EXPRESSION IN MUCOEPIDERMOID CARCINOMA

Resumo: Objective: To evaluate the expression of miR-106a, miR-106b, miR-374-5p, and miR-484 in mucoepidermoid carcinoma (MEC). These microRNAs have been reported in the literature as associated with tumor progression and metastatic potential in different tumors and have been identified as differentially expressed between non-metastatic and metastatic MECs in a previous study from our group. Study Design: Total RNA was extracted from 11 fresh frozen MEC samples and 7 non-neoplastic salivary gland samples. After conversion to cDNA using stem-loop primers, the expression of microRNAs was analyzed by real-time RT-PCR. Expression was normalized to miR-27b and compared to the expression observed in a pool of non-neoplastic salivary gland samples. Results: miR-484 expression was upregulated in 54.5% of samples, representing the most frequently upregulated microRNA. miR-374-5p showed upregulation in 27.3% and downregulation in 18.2%. miR-106b was upregulated in 18.2%, while miR-106a showed both upregulation and downregulation in 9.1% each, indicating heterogeneous expression patterns. Conclusion: miR-484 emerged as the most upregulated microRNA, suggesting a potential role in MEC tumor progression. The remaining miRNAs showed variable expression, indicating context-dependent regulatory mechanisms. These findings suggest the potential of miRNAs as molecular biomarkers and possible therapeutic targets in MEC. Supported by FAPESP (22/14253-7) and CNPq.
